MOLECULAR BIOLOGY

The initiation of molecular biology laboratory at ICH was in 2018. Polymerase Chain Reaction (PCR) instruments were set up. Both PCR and RT-PCR instruments and accessories like Gel electrophoresis, UV Transilluminator and different reagents needed for DNA and RNA related work were arranged. As service for clinical diagnosis, different pathogenic and genetic diseases are detected by fluorescence in real time or by gel electrophoresis of the amplified products. A panel of 22 pathogens for respiratory, gastro and meningo infections can be detected by multiplex RT-PCR instrument of Qiagen. This is being offered for patients both in outdoor and indoor wards. Skin diseases due to fungal infections, Scrub typhus and Pertussis for whooping cough are detected by RT-PCR technique. Among genetic diseases, HLA B-27 allele which is associated with Ankylosing spondilitis is detected as well as cystic fibrosis caused by the major mutation DeltaF 508 in CFTR gene. The different mutations in beta thalassemia are being studied as research project using ARMS-PCR which involves gel electrophoresis following PCR amplification. Another ongoing project is mapping the different mutations in CFTR using sequencing of the whole gene. We plan to detect other infectious and genetic diseases using molecular techniques. Six post-graduate students carried out summer projects in this lab in last three years. We have applied for NABL accreditation of the molecular biology laboratory with HCV as the starting parameter. Later other viral and bacterial pathogens would be diagnosed in the laboratory.
